Study Description:

Our federally funded ADRC recruits and follows cognitively normal, MCI and demented elderly. Our program focuses on AD but we also have small cohorts of DLB, Parkinson disease and early onset AD cases.

Subjects:

Three hundred and forty-one subjects are currently active, with 19% Hispanic and 7.0% African American participants. Table 1 shows the breakdown by ethnicity and diagnosis.

Material Available:

1.  Genomic DNA – 286 subjects

2.  Peripheral blood mRNA – these are serial samples from on ongoing longitudinal study (ImaGene).  Every enrolled subject will have at the end of the study 5 yearly mRNA samples. The baseline diagnoses of our ImaGene subjects are 50 NC, 110 MCI, 22 Parkinson’s disease.

  • 34 with only baseline mRNA
  • 55 with BL and mo12 mRNA
  • 32 with BL+mo12+mo24 mRNA
  • 54 with BL+mo12+mo24+mo36 mRNA
  • 7 with BL+mo12+mo24+mo36+mo48 mRNA

 

3.  Plasma/serum –  collected longitudinally  as well. The baseline diagnoses are 67 NC, 106 MCI , 28 AD, 22 Parkinson’s disease, and 11 other.

  • 84 with only baseline
  • 54 with BL and mo12
  • 35 with BL+mo12+mo24
  • 53 with BL+mo12+mo24+mo36
  • 8 with BL+mo12+mo24+mo36+mo48.

 

4.  CSF – 15 samples total (1 NC, 3AD, 4 MCI, 7 other)

Collection and Storage Methods:

PLASMA AND SERUM

1.  Peripheral blood is collected in  purple  tubes

 a. Samples are centrifuged at 3000 rpm for 15 minutes at room temperature.

 b. Plasma is stored as  plasma in each cryovial.
 c.  Wyeth inhibitor is added to at least 2 aliquots.

2.  After  30 min wait, begin SERUMprep (gold tube).

 a. Serum samples are centrifuged at 3000rpm for 15 min at room temperatue.

 b. Samples are aliquotted into labeled small nalgenes, 0.5 in each.

 c.  Wyeth inhibitor is added to 1-2 aliquots.

3.  Store both plasma and serum @ -80ºC; log into spreadsheet

DNA AND RNA

Peripheral blood samples are drawn in two PAXgene tubes. Total RNA is extracted using the PAXgene blood RNA kit (Qiagen).  Genomic DNA is extracted using standard procedures.

Other Measurements available for this cohort:

Structural MRI - We have at least one MRI image on 190 subjects.

ImaGene subjects are being followed and will ultimately have serial yearly MRIs (5 scans 12 months apart), yearly mRNA (5 samples 12 months apart), yearly plasma/serum (5 samples, 12 months apart) and have all provided genomic DNA.
