New probabilistic methods for DTI evaluations of nerve fiber bundles

Norbert Schuff, Ph.D.
Adjunct Professor, University of California San Francisco
Center for Imaging of Neurodegenerative Diseases

Abstract:
Diffusion tensor imaging (DTI) provides unique information about the microstructure of white matter. However, the best approach to maximize information from DTI is still a matter of intense debate, especially when it comes to quantifying the integrity of nerve fiber bundles,. Recently, we explored the value of information theoretic measures, such as entropy and Kullback-Leibler divergence, to quantify DTI variations along fiber structures as index for fiber integrity within a probabilistic framework. Simulations suggest that the approach is useful for capturing global and local features of the fibers. We also applied the approach to experimental DTI data from patients with cognitive deficits, including Alzheimer’s disease (AD). We found that the results are consistent with the idea that cognitive impairment toward AD is associated with diminishing features of fiber bundles.
